NM_002878.4(RAD51D):c.43G>A (p.Glu15Lys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E15K variant (also known as c.43G>A), located in coding exon 1 of the RAD51D gene, results from a G to A substitution at nucleotide position 43. The glutamic acid at codon 15 is replaced by lysine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
