NM_007294.4(BRCA1):c.4344C>G (p.Ser1448Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S1448R variant (also known as c.4344C>G), located in coding exon 11 of the BRCA1 gene, results from a C to G substitution at nucleotide position 4344. The serine at codon 1448 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
