Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000075.4(CDK4):c.421C>G (p.Leu141Val), citing Ambry Variant Classification Scheme 2023: The p.L141V variant (also known as c.421C>G), located in coding exon 3 of the CDK4 gene, results from a C to G substitution at nucleotide position 421. The leucine at codon 141 is replaced by valine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
