NM_000057.4(BLM):c.4154C>T (p.Thr1385Ile) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T1385I variant (also known as c.4154C>T), located in coding exon 21 of the BLM gene, results from a C to T substitution at nucleotide position 4154. The threonine at codon 1385 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
