NM_000492.4(CFTR):c.4151T>C (p.Ile1384Thr) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I1384T variant (also known as c.4151T>C), located in coding exon 26 of the CFTR gene, results from a T to C substitution at nucleotide position 4151. The isoleucine at codon 1384 is replaced by threonine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
