Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003072.5(SMARCA4):c.4022A>G (p.Asp1341Gly), citing Ambry Variant Classification Scheme 2023: The p.D1341G variant (also known as c.4022A>G), located in coding exon 28 of the SMARCA4 gene, results from an A to G substitution at nucleotide position 4022. The aspartic acid at codon 1341 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
