NM_007294.4(BRCA1):c.3979C>G (p.Gln1327Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q1327E variant (also known as c.3979C>G), located in coding exon 9 of the BRCA1 gene, results from a C to G substitution at nucleotide position 3979. The glutamine at codon 1327 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
