NM_000321.3(RB1):c.395T>A (p.Phe132Tyr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the RB1 gene (transcript NM_000321.3) at coding-DNA position 395, where T is replaced by A; at the protein level this means replaces phenylalanine at residue 132 with tyrosine — a missense variant. Submitter rationale: The p.F132Y variant (also known as c.395T>A), located in coding exon 4 of the RB1 gene, results from a T to A substitution at nucleotide position 395. The phenylalanine at codon 132 is replaced by tyrosine, an amino acid with highly similar properties. This alteration was identified in 0/1358 non-cancer control individuals and in 1/57 cases, in a study looking at cancer predisposition mutations in patients with cutaneous melanoma and a history of at least two additional non-cutaneous melanoma primary cancers (Pritchard AL et al. PLoS One, 2018 Apr;13:e0194098). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
