Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003000.3(SDHB):c.367A>G (p.Lys123Glu), citing Ambry Variant Classification Scheme 2023: The p.K123E variant (also known as c.367A>G), located in coding exon 4 of the SDHB gene, results from an A to G substitution at nucleotide position 367. The lysine at codon 123 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
