NM_000057.4(BLM):c.3668C>A (p.Thr1223Lys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T1223K variant (also known as c.3668C>A), located in coding exon 18 of the BLM gene, results from a C to A substitution at nucleotide position 3668. The threonine at codon 1223 is replaced by lysine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
