Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000179.3(MSH6):c.3623C>G (p.Ser1208Cys), citing Ambry Variant Classification Scheme 2023: The p.S1208C variant (also known as c.3623C>G), located in coding exon 7 of the MSH6 gene, results from a C to G substitution at nucleotide position 3623. The serine at codon 1208 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
