Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.3529G>T (p.Gly1177Cys), citing Ambry Variant Classification Scheme 2023: The p.G1177C variant (also known as c.3529G>T), located in coding exon 20 of the DICER1 gene, results from a G to T substitution at nucleotide position 3529. The glycine at codon 1177 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
