Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002878.4(RAD51D):c.34C>T (p.Leu12Phe), citing Ambry Variant Classification Scheme 2023: The p.L12F variant (also known as c.34C>T), located in coding exon 1 of the RAD51D gene, results from a C to T substitution at nucleotide position 34. The leucine at codon 12 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
