This variant lies in the ALK gene (transcript NM_004304.5) at 4 bases into the intron immediately after coding-DNA position 3359, where A is replaced by G. Submitter rationale: The ALK c.3359+4A>G intronic change results in an A to G substitution at the +4 position of intron 20 of the ALK gene. Algorithms that predict the impact of sequence changes on splicing indicate that this change may result in the activation of a cryptic donor site, however RNA data is not available to confirm this prediction. This variant is absent in gnomAD v2.1.1 (https://gnomad.broadinstitute.org/). To our knowledge, this variant has not been reported in the literature in individuals with ALK-related neuroblastic tumor susceptibility. In summary, the evidence currently available is insufficient to determine the clinical significance of this variant. It has therefore been classified as of uncertain significance.
