NM_004304.5(ALK):c.3346A>C (p.Ile1116Leu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I1116L variant (also known as c.3346A>C), located in coding exon 20 of the ALK gene, results from an A to C substitution at nucleotide position 3346. The isoleucine at codon 1116 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
