Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.3335C>T (p.Pro1112Leu), citing Ambry Variant Classification Scheme 2023: The p.P1112L variant (also known as c.3335C>T), located in coding exon 20 of the ALK gene, results from a C to T substitution at nucleotide position 3335. The proline at codon 1112 is replaced by leucine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
