Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007294.4(BRCA1):c.3331C>G (p.Gln1111Glu), citing Ambry Variant Classification Scheme 2023: The p.Q1111E variant (also known as c.3331C>G), located in coding exon 9 of the BRCA1 gene, results from a C to G substitution at nucleotide position 3331. The glutamine at codon 1111 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
