Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000368.5(TSC1):c.3303G>C (p.Glu1101Asp), citing Ambry Variant Classification Scheme 2023: The p.E1101D variant (also known as c.3303G>C), located in coding exon 21 of the TSC1 gene, results from a G to C substitution at nucleotide position 3303. The glutamic acid at codon 1101 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
