Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.3275+5G>A, citing Ambry Variant Classification Scheme 2023: The c.3275+5G>A intronic variant results from a G to A substitution 5 nucleotides after coding exon 26 in the POLE gene. This nucleotide position is not well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site and will result in the creation or strengthening of a novel splice donor site. Based on the available evidence, the clinical significance of this variant remains unclear.
