NM_005732.4(RAD50):c.95C>A (p.Thr32Lys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T32K variant (also known as c.95C>A), located in coding exon 1 of the RAD50 gene, results from a C to A substitution at nucleotide position 95. The threonine at codon 32 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
