Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_024675.4(PALB2):c.3269T>G (p.Phe1090Cys), citing Ambry Variant Classification Scheme 2023: The p.F1090C variant (also known as c.3269T>G), located in coding exon 12 of the PALB2 gene, results from a T to G substitution at nucleotide position 3269. The phenylalanine at codon 1090 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
