Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000057.4(BLM):c.3262G>C (p.Val1088Leu), citing Ambry Variant Classification Scheme 2023: The p.V1088L variant (also known as c.3262G>C), located in coding exon 16 of the BLM gene, results from a G to C substitution at nucleotide position 3262. The valine at codon 1088 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
