Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000051.4(ATM):c.3231T>A (p.Phe1077Leu), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 3231, where T is replaced by A; at the protein level this means replaces phenylalanine at residue 1077 with leucine — a missense variant. Submitter rationale: Variant summary: ATM c.3231T>A (p.Phe1077Leu) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 251296 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.3231T>A in individuals affected with Ataxia-Telangiectasia and no experimental evidence demonstrating its impact on protein function have been reported. Two clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. All laboratories classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
