Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000179.3(MSH6):c.308A>T (p.Tyr103Phe), citing Ambry Variant Classification Scheme 2023. This variant lies in the MSH6 gene (transcript NM_000179.3) at coding-DNA position 308, where A is replaced by T; at the protein level this means replaces tyrosine at residue 103 with phenylalanine — a missense variant. Submitter rationale: The p.Y103F variant (also known as c.308A>T), located in coding exon 2 of the MSH6 gene, results from an A to T substitution at nucleotide position 308. The tyrosine at codon 103 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. In addition, the CoDP in silico tool predicts this alteration to have minor impact on molecular function, with a score of 0.001 (Terui H et al. J. Biomed. Sci. 2013 Apr;20:25). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
