NM_000057.4(BLM):c.3005A>C (p.Lys1002Thr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K1002T variant (also known as c.3005A>C), located in coding exon 14 of the BLM gene, results from an A to C substitution at nucleotide position 3005. The lysine at codon 1002 is replaced by threonine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
