NM_032043.3(BRIP1):c.854A>C (p.His285Pro) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H285P variant (also known as c.854A>C), located in coding exon 6 of the BRIP1 gene, results from an A to C substitution at nucleotide position 854. The histidine at codon 285 is replaced by proline, an amino acid with similar properties. This amino acid position is highly conserved through mammals but not in all available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
