NM_007294.4(BRCA1):c.1052A>T (p.Lys351Ile) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K351I variant (also known as c.1052A>T), located in coding exon 9 of the BRCA1 gene, results from an A to T substitution at nucleotide position 1052. The lysine at codon 351 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
