NM_000548.5(TSC2):c.1014C>G (p.Ile338Met) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I338M variant (also known as c.1014C>G), located in coding exon 10 of the TSC2 gene, results from a C to G substitution at nucleotide position 1014. The isoleucine at codon 338 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
