Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000057.4(BLM):c.2686C>T (p.Leu896Phe), citing Ambry Variant Classification Scheme 2023: The p.L896F variant (also known as c.2686C>T), located in coding exon 13 of the BLM gene, results from a C to T substitution at nucleotide position 2686. The leucine at codon 896 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
