Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_000038.6(APC):c.252A>G (p.Gly84=), citing Sema4 Curation Guidelines: The APC c.252A>G (p.G84=) variant has not been reported in the literature to our knowledge. It was observed in 3/113730 chromosomes of the European (non-Finnish) subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). This variant has been reported in ClinVar (Variation ID: 821433). In silico tools suggest that the variant may create or strengthen a cryptic splice site, though these predictions have not been confirmed by functional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
