Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000251.3(MSH2):c.2427G>C (p.Glu809Asp), citing Ambry Variant Classification Scheme 2023: The p.E809D variant (also known as c.2427G>C), located in coding exon 14 of the MSH2 gene, results from a G to C substitution at nucleotide position 2427. The glutamic acid at codon 809 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
