NM_000388.4(CASR):c.2314G>A (p.Ala772Thr) was classified as Uncertain significance for Nephrolithiasis/nephrocalcinosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A772T variant (also known as c.2314G>A), located in coding exon 6 of the CASR gene, results from a G to A substitution at nucleotide position 2314. The alanine at codon 772 is replaced by threonine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. In addition, the evidence for the gene-disease relationship is limited for pancreatitis and cancer predisposition; therefore, the clinical significance of this variant for CASR-related pancreatitis and cancer predisposition is unclear. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
