NM_000179.3(MSH6):c.224G>C (p.Gly75Ala) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G75A variant (also known as c.224G>C), located in coding exon 1 of the MSH6 gene, results from a G to C substitution at nucleotide position 224. The glycine at codon 75 is replaced by alanine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
