NM_001042492.3(NF1):c.2132G>A (p.Arg711His) was classified as Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R711H variant (also known as c.2132G>A), located in coding exon 18 of the NF1 gene, results from a G to A substitution at nucleotide position 2132. The arginine at codon 711 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
