Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000249.4(MLH1):c.2162A>C (p.Tyr721Ser), citing Ambry Variant Classification Scheme 2023: The p.Y721S variant (also known as c.2162A>C), located in coding exon 19 of the MLH1 gene, results from an A to C substitution at nucleotide position 2162. The tyrosine at codon 721 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
