Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004656.4(BAP1):c.2065G>C (p.Ala689Pro), citing Ambry Variant Classification Scheme 2023: The p.A689P variant (also known as c.2065G>C), located in coding exon 17 of the BAP1 gene, results from a G to C substitution at nucleotide position 2065. The alanine at codon 689 is replaced by proline, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
