NM_000535.7(PMS2):c.2025A>T (p.Glu675Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E675D variant (also known as c.2025A>T), located in coding exon 12 of the PMS2 gene, results from an A to T substitution at nucleotide position 2025. The glutamic acid at codon 675 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
