NM_001042492.3(NF1):c.1859G>A (p.Ser620Asn) was classified as Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S620N variant (also known as c.1859G>A), located in coding exon 17 of the NF1 gene, results from a G to A substitution at nucleotide position 1859. The serine at codon 620 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
