NM_177438.3(DICER1):c.1859A>G (p.Asp620Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D620G variant (also known as c.1859A>G), located in coding exon 10 of the DICER1 gene, results from an A to G substitution at nucleotide position 1859. The aspartic acid at codon 620 is replaced by glycine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
