Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000264.5(PTCH1):c.1828A>G (p.Ile610Val), citing Ambry Variant Classification Scheme 2023: The p.I610V variant (also known as c.1828A>G), located in coding exon 13 of the PTCH1 gene, results from an A to G substitution at nucleotide position 1828. The isoleucine at codon 610 is replaced by valine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
