Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002485.5(NBN):c.1785G>C (p.Met595Ile), citing Ambry Variant Classification Scheme 2023: The p.M595I variant (also known as c.1785G>C), located in coding exon 11 of the NBN gene, results from a G to C substitution at nucleotide position 1785. The methionine at codon 595 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
