Uncertain significance for Cystic fibrosis — the classification assigned by Ambry Genetics to NM_000492.4(CFTR):c.178G>C (p.Glu60Gln), citing Ambry Variant Classification Scheme 2023: The p.E60Q variant (also known as c.178G>C), located in coding exon 3 of the CFTR gene, results from a G to C substitution at nucleotide position 178. The glutamic acid at codon 60 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
