Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002485.5(NBN):c.1730A>G (p.Asp577Gly), citing Ambry Variant Classification Scheme 2023: The p.D577G variant (also known as c.1730A>G), located in coding exon 11 of the NBN gene, results from an A to G substitution at nucleotide position 1730. The aspartic acid at codon 577 is replaced by glycine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
