NM_005732.4(RAD50):c.1717A>C (p.Lys573Gln) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K573Q variant (also known as c.1717A>C), located in coding exon 11 of the RAD50 gene, results from an A to C substitution at nucleotide position 1717. The lysine at codon 573 is replaced by glutamine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
