Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_024675.4(PALB2):c.1712A>T (p.Asp571Val), citing Ambry Variant Classification Scheme 2023: The p.D571V variant (also known as c.1712A>T), located in coding exon 5 of the PALB2 gene, results from an A to T substitution at nucleotide position 1712. The aspartic acid at codon 571 is replaced by valine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
