NM_007194.4(CHEK2):c.1503G>T (p.Glu501Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E501D variant (also known as c.1503G>T), located in coding exon 13 of the CHEK2 gene, results from a G to T substitution at nucleotide position 1503. The glutamic acid at codon 501 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
