Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002528.7(NTHL1):c.-11A>T, citing Ambry Variant Classification Scheme 2023: The p.Q5L variant (also known as c.14A>T), located in coding exon 1 of the NTHL1 gene, results from an A to T substitution at nucleotide position 14. The glutamine at codon 5 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
