Submitter rationale: The p.A49T variant (also known as c.145G>A), located in coding exon 2 of the PMS2 gene, results from a G to A substitution at nucleotide position 145. The alanine at codon 49 is replaced by threonine, an amino acid with similar properties. This variant was detected in an Italian cohort of 113 non-BRCA patients with a personal and/or familial history of breast cancer, ovarian cancer, and/or pancreatic cancer (Germani A et al. J Clin Med, 2020 Sep;9:). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
