NM_007194.4(CHEK2):c.13T>C (p.Ser5Pro) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S5P variant (also known as c.13T>C), located in coding exon 1 of the CHEK2 gene, results from a T to C substitution at nucleotide position 13. The serine at codon 5 is replaced by proline, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
