NM_007294.4(BRCA1):c.135-4A>G was classified as Uncertain significance by Quest Diagnostics Nichols Institute San Juan Capistrano, citing Quest Diagnostics criteria. This variant lies in the BRCA1 gene (transcript NM_007294.4) at 4 bases into the intron immediately before coding-DNA position 135, where A is replaced by G. Submitter rationale: The BRCA1 c.135-4A>G variant has not been reported in individuals with BRCA1-related conditions in the published literature. One in vitro study of haploid cell survival using saturated genome editing shows that this variant retains BRCA1 functional activity (PMID: 30209399 (2018)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Analysis of this variant using software algorithms for the prediction of the effect of nucleotide changes on splicing yielded predictions that this variant does not affect BRCA1 mRNA splicing. Based on the available information, we are unable to determine the clinical significance of this variant.
